Scalable and sustainable synthesis

Fe(II)- and α-ketoglutarate-dependent oxygenases (KGOs), also known as 2-oxoglutarate-dependent oxygenases (2OGs) or αKGDs, form a large and diverse family of non-heme iron enzymes that catalyze oxidative transformations using molecular oxygen and α-ketoglutarate (αKG) as co-substrate. Unlike complex P450 enzymes, KGOs are streamlined and efficient, with no intricate domain structures that can lead to instability. This means they are robust under diverse conditions, making them ideal for reliable, scalable applications in even the most demanding workflows.

KGOs, often derived from bacterial sources, are expressed intracellularly, where they influence native metabolic pathways by metabolite modification. These enzymes are renowned for catalyzing a variety of reactions, most prominently hydroxylation, but also halogenation, desaturation, and ring formation, all under mild aqueous conditions at ambient temperature and pressure. These enzymes only require α-ketoglutarate as oxygen donor which can be continuously supplemented or regenerated during the reaction via an enzymatic side reaction. This cofactor-based mechanism confers high stereo-, chemo-, and regioselectivity, enabling transformations that are chemically challenging through conventional synthesis.

Industrially, the scalability of KGO-catalyzed reactions has been demonstrated, with fermentation-based processes now capable of yielding up to 400 kilograms enzyme. Such scalability, combined with their operational mildness and catalytic precision, establishes KGOs as highly valuable biocatalysts for sustainable synthesis of fine chemicals, pharmaceuticals, and complex intermediates.

Curated enzyme panels

Identifying the right Fe(II)-α-ketoglutarate oxygenase for a given substrate remains a major challenge in biocatalytic research, where experimental screening often consumes time, material, and cost. Conventional enzyme panels frequently yield few or no active hits due to the limited substrate scope or incomplete enzyme representation.

Curated KGO kits address this gap by leveraging computational curation and predictive modeling to match enzyme reactivity with specific substrate classes. By integrating sequence homology searches, structural modeling, and active-site profiling, in silico selection enables the rational assembly of focused enzyme panels with improved hit probability. This approach significantly reduces the cost and duration of experimental discovery, as only the most promising candidates are expressed and tested.

Panels dedicated to particular substrate categories, such as amino acids, nucleic acids, or heterocyclic scaffolds, allow rapid identification of functional biocatalysts and facilitate reaction optimization in early development. Curated kits thus streamline biocatalyst discovery while maintaining high predictive accuracy, accelerating the path from screening to scalable synthesis.
